摘要:
目的 探讨红景天苷对家兔50%减体积原位肝移植实验性术后疲劳综合征(POFs)的作用及其机制。
方法 将36只家兔随机分为3组,每组12只。A组不手术,B、C组先采用小肠切除-吻合术建立POFs模型,造模第10天将两组肝脏进行交叉50%减体积原位移植,术后C组用10 g/L红景天苷按2 mL/kg剂量经尾静脉注射治疗7 d;A组、B组按1 mL/kg剂量尾静脉注射生理盐水。检测3组家兔造模前、肝移植术后24 h和治疗第7天大脑皮质电位、外周血5-羟色胺(5-HT)和垂体生长激素(GH)水平,并检测谷丙转氨酶(ALT)、谷草转氨酶(AST)和总胆红素(TBIL)含量。
结果 C组家兔治疗第7天与肝移植术后24 h及B组比较,5-HT、大脑皮质电位及肝功能指标ALT、AST和TBIL均明显降低,GH明显增高,差异均有显著意义(F=50.21~102.75,q=3.98~14.02,P<0.05)。
结论 红景天苷可降低家兔50%减体积原位肝移植实验性POFs并促进肝功能恢复,对家兔实验性POFs有一定程度的防治作用。
Abstract:
Objective  To investigate the effect of salidroside on postoperative fatigue syndrome (POFs) in rabbits undergoing 50% reduced-size orthotopic liver transplantation.
Methods  A total of 36 rabbits were randomly divided into three groups, with 12 rabbits in each group. Group A was not given surgery; in groups B and C, small intestinal resection-anastomosis was performed to establish a rabbit model of POFs, and 50% reduced-size orthotopic liver transplantation was performed on day 10 after modeling. After surgery, the rabbits in group C were given tail vein injection of 10 g/L salidroside at a dose of 2 mL/kg for 7 days, and those in groups A and B were given tail vein injection of normal saline at a dose of 1 mL/kg. Cerebral cortex potentials, peripheral 5-hydroxytryptamine (5-HT) and pituitary growth hormone (PGH) levels, and levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and total bilirubin (TBIL) were measured before modeling, at 24 h after liver transplantation, and on day 7 of treatment.
Results  Group C had significant reductions in 5-HT, cerebral cortex potentials, and le-
vels of ALT, AST, and TBIL and a significant increase in PGH at 24 h after liver transplantation and on day 7 of treatment, and compared with group B, group C had significantly lower 5-HT, cerebral cortex potentials, and levels of ALT, AST, and TBIL and a significantly higher level of PGH (F=50.21-102.75,q=3.98-14.02,P<0.05).
Conclusion  In rabbits undergoing 50% reduced-size orthotopic liver transplantation, salidroside can reduce experimental POFs and promote the recovery of liver function, and therefore, it can be used for the prevention and treatment of experimental POFs in rabbits.
