摘要:
 目的 观察大鼠主动脉球囊损伤后血管组织中Apelin的表达变化及缬沙坦治疗对其表达的影响,探讨缬沙坦在预防再狭窄中的作用及其机制。
方法 将48只Wistar大鼠随机分为对照组、手术组及药物组,各16只。手术组和药物组大鼠用自制的2F球囊导管进行主动脉内皮损伤,构建大鼠主动脉内皮球囊损伤模型;对照组不行主动脉球囊损伤。药物组术前1 d~术后14 d(或28 d)给予缬沙坦(20 mg/(kg?d))进行干预。各组分别在术后14、28 d取主动脉组织,应用苏木精-伊红染色观察血管内膜的变化,实时荧光定量逆转录聚合酶链反应技术、免疫组织化学方法检测主动脉组织Apelin的mRNA和蛋白表达水平。
结果 术后14、28 d手术组血管内膜较对照组显著增厚,药物组内膜厚度较手术组明显降低,差异有显著性(F=168.85、233.62,P<0.05)。手术组内皮损伤后14、28 d主动脉组织Apelin mRNA和蛋白表达较对照组显著升高,药物组Apelin mRNA和蛋白表达较手术组明显增加,差异有显著性(F=26.74~48.71,P<0.05)。
结论 大鼠主动脉球囊损伤后血管组织中Apelin的表达升高,缬沙坦抑制主动脉球囊损伤后内膜增生可能与其上调Apelin的表达有关。
Abstract:
 Objective To observe the changes in vascular expression of Apelin after aortic balloon injury in rats and the effects of valsartan treatment on Apelin expression, and to investigate the role of valsartan in the prevention of restenosis and its mechanism.
Methods Forty-eight Wistar rats were randomly and equally divided into control group, operation group, and drug group. To establish a rat model of aortic endothelial balloon injury, self-made 2F balloon catheters were used to induce aortic endothelial injury in the operation group and the drug group but not the control group. The drug group was given valsartan (20 mg/(kg?d)) from 1 d before operation to 14 (or 28) d after operation. The aorta was collected at 14 and 28 d after operation. The changes in intima-media thickness were observed by HE staining. The mRNA and protein expression of Apelin in the aorta was measured by RT-PCR and immunohistochemistry, respectively.
Results At 14 and 28 d after operation, the operation group had significantly increased intima-media thickness compared with the control group, while the drug group had significantly reduced intima-media thickness compared with the operation group (F=168.85,233.62;P<0.05); the operation group had significantly higher mRNA and protein expression of Apelin than the control group, while the drug group had significantly higher mRNA and protein expression of Apelin than the operation group (F=26.74-48.71,P<0.05).
Conclusion After aortic balloon injury, the vascular expression of Apelin increases in rats. Valsartan inhibits intimal hyperplasia after aortic balloon injury, probably by upre-
gulating the expression of Apelin.
