摘要:
目的 探讨类赖氨酰氧化酶1(LOXL1)基因rs1048661位点多态性与剥脱综合征(XFS)和剥脱综合征性青光眼(XFG)发病风险的相关性。
方法 计算机检索PubMed、Web of science、Embase、万方、中国期刊全文数据库(CNKI)、维普数据库,以“类赖氨酰氧化酶1基因;剥脱综合征;剥脱综合征性青光眼;多态性,单核苷酸;Meta分析;lysyl oxidase-like 1;Exfoliation syndrome;Exfoliation glaucoma;polymorphism;single nucleotide;Meta-analysis”为关键词,查找国内外关于LOXL1基因rs1048661位点多态性与XFS和XFG易感性关系的病例-对照研究,检索时限为建库至2017年5月。由2名独立的评价者根据纳入及排除标准筛查文献,提取相关资料后,运用Stata 10.0软件进行Meta分析。
结果 最终纳入36篇文献44个研究,其中包括病例组病人4 910例,健康对照组8 443例,经异质性检验,各研究间存在着异质性(I2=92.7%,P<0.001)。采用随机效应模型对数据进行Meta分析,结果显示,对于总体人群而言LOXL1基因rs1048661位点多态性与XFS/XFG易感性关联无统计学意义(P>0.05)。但根据人群种族进行亚组分析显示,在高加索人群中,LOXL1基因rs1048661位点多态性可增加XFS/XFG易感性;亚洲人群中LOXL1基因rs1048661位点多态性可降低XFS/XFG易感性;在非洲人群中,LOXL1基因rs1048661位点多态性与XFS/XFG易感性关联无统计学意义。在研究疾病种类基础上进行亚组分析显示,XFS病人中LOXL1基因rs1048661位点多态性可增加XFS的易感性;在XFG病人中,LOXL1基因rs1048661位点多态性可增加XFG的易感性。
结论 LOXL1基因rs1048661位点多态性可降低亚洲人群XFS/XFG易感性,增加高加索人群XFS/XFG易感性;但在非洲人群中该基因多态性与XFS/XFG易感性无统计学关联。
Abstract:
Objective To investigate the association of lysyl oxidase-like 1 (LOXL1) rs1048661 polymorphisms with the risk of exfoliation syndrome (XFS) and exfoliation glaucoma (XFG).
Methods PubMed, Web of Science, Embase, Wanfang Data, CNKI, and VIP were searched for case-control studies on the association of LOXL1 rs1048661 polymorphisms with the risk of XFS and XFG in China and foreign countries published up to May 2017, with the key words of lysyl oxidase-like 1, exfoliation syndrome, exfoliation glaucoma, polymorphism, single nucleotide, and Meta-analysis in both Chinese and English languages. Two reviewers independently screened the articles according to inclusion and exclusion criteria, and after related data were extracted, Stata 10.0 software was used to perform the Meta-analysis.
Results A total of 36 articles with 44 studies were enrolled, with 4 910 patients in case group and 8 443 persons in healthy control group. The test for heterogeneity showed that there was heterogeneity between these studies (I2=92.7%,P<0.001). The random effect model was used for the Meta-analysis of related data, and the results showed that there was no significant association between LOXL1 rs1048661 polymorphisms and susceptibility to XFS/XFG in the overall population. According to the subgroup analysis based on ethnicity, in the Caucasian population, LOXL1 rs1048661 polymorphism increased the susceptibility to XFS/XFG; in the Asian population, LOXL1 rs1048661 polymorphism reduced the susceptibility to XFS/XFG; in the African population, there was no significant association between LOXL1 rs1048661 polymorphisms and susceptibility to XFS/XFG. According to the subgroup analysis based on disease type, in patients with XFS, LOXL1 rs1048661 polymorphism increased the susceptibility to XFS; in patients with XFG, LOXL1 rs1048661 polymorphism increased the susceptibility to XFG.
Conclusion LOXL1 rs1048661 polymorphism can reduce the susceptibility to XFS/XFG in the Asian population and increase the susceptibility to XFS/XFG in Caucasians, and there is no significant association between LOXL1 rs1048661 polymorphism and the susceptibility to XFS/XFG in the African population.
